鸭淋巴细胞转化试验(MTT法)最佳条件的研究

摘　　要：通过对培养温度、小牛血清及刀豆蛋白A(ConA)的浓度以及培养时间 4个参数的研究 ,确定了鸭淋巴细胞转化试验 (MTT法 )的最优培养条件。结果表明 ,采用 3 9.0℃、含 1 0 %小牛血清和 5μg/mLConA的RPMI1 64 0培养 66h可获得最大的OD570nm光吸值。Through studying four parameters of cultural temperature and time, density of bovine serum and ConA, the optimum cultural condition of duck's proliferation of lymphocyte is ascertained. The results indicate that the maximum absorbance of 570nm test wavelength can be acquired after 66 hours under the condition of 39.0℃, RPMI1640 medium containing 10 percent of bovine serum and 5μg/mL ConA.
