摘 要:采用原生质体融合技术 ,将两株具有含氯有机化合物降解特性的菌 - SY- 1和 RB- 1融合构建成一株高效降解含氯有机化合物的工程菌。实验得出 :当溶菌酶浓度为 1.5 mg/ml,酶解 6 0 min时 ,假单胞菌和诺卡氏菌的原生质体形成率分别为 33.3%和 44 .4% ;在 30 % PEG6 0 0 0助融下 10 min二者的融合率为 0 .3%。应用于造纸漂白废水 ,融合菌去除CODCr和 TCL的能力比混合菌分别提高了 72 .0 5 %和 190 %By the technique of protoplast fusion,interspecific fusants between Pseudomanos SY l and Norcardia RB 1 had been obtained.The superior conditions of protoplast formation,protoplast fusion and wastewater treatment using the fusants were studied.It is better to prepare the protoplast by using lysozyme 1.5mg/ml,10min.The protoplast formation rates of Pseudomamos and Norcardia were 33.3% and 44.4% respectively.The fusion was carried out in NSM containing 30% PEG(MW6000) and the fusion frequency was about 0.3%.At last,the stable fusants were sued to treat the bleaching wastewater.It is obtained that the removal rates of COD Cr and TCL are respectively 72.05% and 190% higher than that of the mixed parent strains.
