摘 要:目的 探讨系统性红斑狼疮(SL)患者外周血淋巴细胞Fas及其相应配体FasL的表达水平及在SLE发病中的作用。方法 应用逆转录-聚合酶链反应(RT-PCR)检测34例活动期SLE患者和30例正常人外周血单一核细胞(PBMC)中 Fas和 FasL mRNA的表达水平。结果活动期 SLE患者 PBMC中 FasL的阳性检出率为 91. 18%,明显高于正常对照组(63.30%),差异非常显著(P<0.01);Fas的阳性检出率与对照组相比无明显差异(P>0.05)。活动期SLE患者.PBMC中Fas和FasL mRNA的平均表达水平均高于正常对照组,差异显著(P<0.05)。结论Fas和FasL的高水平表达可能与SLE患者外周血T淋巴细胞活化、凋亡增加有关,Fas介导的淋巴细胞凋亡可能参与了SLE的发病过程。Objective To investigate the levels of Fas and Fas ligand mRNA expression in the patients with systemic lupus erythematosus. Methods Applying reverse transcription-polymerase reaction(RT-PCR) technique to analyze Fas and FasL mR- NA expression in the peripheral blood mononuclear cells(PBMC) from 34 active SLE patients and 30 cases of healthy subjects. Results The SLE patients with positive rate of FasL expression accounted for 91. 18% of the SLE patients in active stage,com- pared with that of normal controls(63. 30% ),The difference was markedly markedly significant(p< 0. 01). The mean levels of Fas and FasL mRNA expression in active SLE group were increased(p< 0. 05), there was no statistical significance in the positive rate of Fas expression between patients with active SLE and normal controls(P>0. 05). Conclusions The higher expression of Fas and FasL may contribute to the activation of lymphocytes and/or the increased apoptosis of lymphocytes in patients with systemic lu- pus erythematosus. Fas-mediated apoptosis may be involved in the pathogenesis. of SLE.
