摘  要:目的 了解基因芯片技术在筛查正常或异常胚胎发育的相关基因表达变化中的应用价值。方法 用体外全胚胎培养和含有14 000条人类基因的cDNA表达谱芯片,对同型半胱氨酸(0~4mmol/L)诱导的大鼠畸形胚胎进行基因表达谱分析。结果 4mmol/L的同型半胱氨酸(HCY)能诱发体外培养的大鼠胚胎发生畸变,畸形发生率为66.7%(24/36),显著高于对照组(P<0.05);基因表达谱分析发现有18条基因表达明显下调。结论 同型半胱氨酸对早期大鼠胚胎具有致畸作用;运用基因芯片技术能够有效地筛检出新的胚胎发育相关基因。Objective To investigate the role of gene chip technique in screening normal or abnormal developing embryo related genes. Methods The in vitro whole embryo culture (WEC) and expression cDNA microarry which representing a set of 14 000 human genes were used to observe and analysis the gene expression pattern of abnormal developed rat embryos by homocysteine (0-4 mmol/L) induced. Results 4 mmol/L homocysteine could cause dysmorphogenesis of the day 9. 5 embryos cultured in vitro. The embryotic defect rate was 66.7 % (24/36) and higher than that of control group (P<0. 05 ). There were 18 significantly downregulated genes which were identified by using the cDNA microarray. Conclusion Homocysteine may be a developing toxic factor and can induce teratogenesis for early rat embryo. cDNA microarray for gene expression pattern analysis may be a very useful technique to discover and identify novel genes related with embryotic develop
