风疹病毒单克隆抗体的研究

摘　　要：用风疹病毒(RV)Gos-10株免疫BALB/c小鼠,取其脾细胞与Sp2/0细胞融合,用ELISA法筛选分泌RV McAb的杂交瘤细胞6株,经克隆培养后发现其中的1A_9株能稳定分泌高效价特异性McAb,经鉴定为IgG2a,经ELISA检测、抗原吸收试验、中和试验及HI试验征明该McAb是种特异性的,能与各株RV发生交叉反应,而与其它种病毒不发生反应,证明此McAb对RV抗原有高度特异性与敏感性,可用以制备诊断试剂,实现风疹的早期、快速诊断和基础理论研究。This experiment which fused spleen cells of BALB/c mice im-munized by rubella virus Gos-10 strain with Sp2/0 myeloma cells,screened by ELISA has got six hybridomas.The 1A_9 hybridoma which can secrete RV McAb steadly,was cloned for 3 times with limited dilution method and po-sitive rate reached as high as to 100 per cent.This RV McAb was classified as sub-IgG2a.ELISA antigen absorption test,neutralization test,and haemag-glutination inhibition test showed that the McAb is specific and sensitive to RV and can be used to prepare diagnostic kits to diagnose RV infection early and rapidly.
