摘 要:目的 探讨白细胞提取物抗肺炎、肺纤维化的效果及其作用机制。方法 用博来霉素(BLM A6 ) 6mg/kg气管内注入 ,诱生小鼠肺炎和肺纤维化 ;预防治疗组肌内注射白细胞提取物 ,每日5 0 0 μg/kg;阳性对照组肌内注射丹参提取物 (IH76 4 3) ,每日 2 0mg/kg ,对照组肌内注射等体积的生理盐水。 30d后处死动物 ,用电镜观察其病理改变。结果 预防治疗组和阳性对照组经处理后 ,分别有6 0 %和 87%的动物肺组织炎症和纤维化得到改善。电镜观察显示 ,两组大多数动物肺胶原形成细胞数量、炎性细胞渗出、胶原纤维和弹力纤维都较阴性对照组明显减少 ,肺泡毛细血管形态较正常 ,肺泡隔内胶原纤维明显减少 ,保留了肺的蜂窝状结构。结论 白细胞提取物对博来霉素诱致的肺炎、肺纤维化具有一定的防治效果 。Objective To explore the effect of leukocyte extract(LE) on experimental pneumonia and pulmonary fibrosis and its mechanism. Methods Bleomycin(BLM A6) 6 mg/kg was used to induce pneumonia and pulmonary fibrosis in mice by single intratracheal instillation.The prevention and treatment group(LE group) were treated with LE 500 μg/kg per day(im),the positive control group(Salvia Miltiorhiza extract,IH764 3 group) was treated with IH764 3 20 mg/kg per day(im) and the negative control group with equal volume of normal saline.30 d later,the animals were killed and the pathological changes of lung were examined by electronmicroscope. Results Pulmonary inflammation and fibrosis were improved by 60% and 87% in LE and IH764 3 group respectively.The ultrastructure in most of them showed lesser collagen forming cells,infiltrating inflammatory cells,as well as collagen and elastic fibers than those in negative control;the morphology of alveolar capillaries was rather normal;the collagen fibers in alveolar septum was obviously reduced and kept in honeycomb shape of pulmonary structure. Conclusion Leukocyte extract has prophylactic and therapeutic effects on pneumonia and pulmonary fibrosis.Its mechanism may be by way of regulating the non specific immunity of the body.
