摘  要:棘托竹荪 (DictyophoraechinovolvataZang ,ZhengetHu)子实体干品经热水抽提 ,乙醇沉淀 ,蛋白酶法和Sevag法去蛋白后得粗多糖DE。DE经乙醇分级沉淀得到三个级分DE1、DE2和DE3。DE2经DEAE SephadexA - 2 5柱进一步纯化得到棘托竹荪多糖纯品DE2 - 2。DE2 - 2经高效液相色谱法测定为均一物质。红外扫描表明 ,其具有典型的多糖吸收峰。纸层析和气相色谱分析结果表明 ,DE2 - 2的单糖组成为 :D -葡萄糖、D -甘露糖、D -半乳糖和L -岩藻糖 ,其摩尔比为 8.68∶1.0 0∶1.85∶0 .74 ,分子量约为 84 0 0 0。经测定 ,DE2 - 2具有一定的抑瘤活性 ,其抑瘤率为38.93%。The crude polysaccharide DE was extracted from dried fruitbodies of Dictyophora echinovolvata by hot water extraction, ethanol precipitation and deproteinization with protease method and Sevag method.Then three fractions DE1, DE2 and DE3 were obtained by fractional precipitation of DE with ethanol. While pure DE2-2 was obtained by further purification of DE2 with DEAE Sephadex A-25 and its homogeneity was verified by HPLC. And typical absorption peaks of polysaccharide were found with IR-scan. It was proved that DE2-2 was composed of D glucose, D mannose, D galactose and L fucose with a molar ratio of 8.68∶1.00∶1.85∶0.74 by paper and gas phase chromatography. The molecular weight of DE2-2 was estimated to be 84000 with an inhibition rate of 38.93% to Sarcoma-180 implanted in mice.
