摘 要:以转化 CMV外壳蛋白基因 ( CP基因 )的线辣椒 ( Capsicum annuum var. longunt)为试材 ,系统研究了 CP基因介导抗病性的组分和表达特点。摩擦接种黄瓜花叶病毒 ( CMV,浓度 2 0μg/ ml)后 ,转基因线辣椒显症延迟 4~ 1 0 d,显症率和严重度大幅减低 ,仅部分植株表现轻度花叶。接种叶片上 CMV侵入位点数目比未转化对照品种减少 6 7.0 %~ 86 .4 %。接种后第 9天 ,转基因株系接种叶中 CMV含量仅为未转化对照的32 .4 %~ 37.3%。病毒由接种叶向邻叶转出始期 ,比未转化对照晚 9~ 1 5h。接种后 3d,转基因株系 1 6— 1 3的未接种叶中未能检出 CMV,而同期未转化对照已有 4片邻叶检出了 CMV。接种后 6 d,1 6— 1 3仅上下位相邻叶检出了 CMV,此时对照已全株带毒。转基因株系接种后 6 d,整株 CMV含量为未转化对照的 4 2 .5%。这表明线辣椒 CP基因介导的抗病性是多组分的 ,包括对病毒的抗侵入。The components and expression of CP MR to CMV in transgenic chili pepper ( Capisicum annuum var. longunt ) with introduced CP genes were studied. The results revealed that the symptom expression in mechanically inoculated CP (+) transgenic plants was delayed in 4~10 d, incidence and severity of CMV infection was greatly reduced with light mosaic in part of plants. The penetration sites of CMV in leaves of CP (+) plants were 67.0%~86.4% lower than that in CP (-) untransformed plants. The virus concentration in CP (+) leaves in 9 d after inoculation was 32.4%~37.3% of that in CP (-) leaves. The initial time of virus movement from inoculated leaf to adjacent leaves in CP (+) plant was 9~15 h later than that in CP (-) plant. While CMV was detected by ELISA test in 4 adjacent leaves or in all of uninoculated leaves of CP (-) plants, respectively, in 3 d or in 6 d after inoculation, no virus was detected in uninoculated leveas of CP (+) plants in 3 d after inoculation and it was detected only in 2 adjacent leaves in 6 d after inoculation. CMV concentration in CP (+) plants was as low as 42.5% of that in CP (-) plants in 6 d after inoculation. It was concluded that CP MR in transgenic chili pepper was multi-component, which could resist to viral penetration, to viral colonization and to viral multiplication and transportation.
