摘 要:目的 :观察编码IL 12的真核表达载体对HBV基因疫苗诱导Babl c小鼠免疫应答的影响。方法 :肌肉注射基因疫苗pCR3.1 S及IL 12的真核表达载体pWRG316 9,ELISA法检测小鼠血清抗HBs ,4h5 1 Cr释放法检测小鼠脾细胞CTL杀伤活性。结果 :免疫 8w后 ,pCR3 1 S及共注射IL 12真核表达载体组血清 45 0nmA值分别为 0 87± 0 1及 1 6 7± 0 15。CTL细胞杀伤活性分别为 5 0 5 %± 6 4%及 73 3%± 8 8% ,两组差异显著 ,CTL细胞杀伤活性在抗CD4单克隆抗体处理脾细胞悬液后无明显改变 ,抗CD8单克隆抗体处理后明显降低。结论 :IL 12的真核表达载体能提高小鼠对DNA疫苗的免疫应答 ,CTL细胞杀伤活性主要由CD8+ 执行。基因疫苗可能用于预防及治疗HBV感染。Objective:To observe the immune responses to gene vaccines coding HBV surface antigen and interleukin 12 in Balb/c(H 2 d) mice.Methods:The immunization was performed by intramuscular injection in this experiment.Anti HBs in serum was detected by ELISA.HBsAg specific cytotoxic T lymphocytes (CTLs) activity was measured by 51 Cr release assay.Results:8 w after immunization,the serum OD value of the mice coinjected IL 12 vaccine (1.64±0.15) is significantly higher than that of mice injected HBV S DNA vaccine (0.87±0.1) only.CTLs activity of the mice injected HBV S DNA vaccine and coinjected IL 12 vaccine were 50.5%±6.4% and 73.3%±8.8% respectively.After adding anti CD4 + monoclonal antibody in spleen cells,CTLs activity of the mice injected HBV S DNA vaccine and coinjeted IL 12 vaccine were 48.3%±5.9% and 75.6%±9.1% respectively.CTLs activity were 10.6%±1.4% and 16.3%±2.3% respectively after adding anti CD8 + monoclonal antibody in spleen cells.Conclusion:The results showed that the DNA vaccine of pCR3.1 S had strong antigenecity in cellular and humoral immunity which can be promoted by vector coding murine IL 12.CTLs activity were performed by CD8 + cells.DNA vaccine against HBV may be useful for both prophylactic and therapeutic purposes.
