用胸导管引流细胞制备人白细胞间素Ⅱ条件的探讨

摘　　要：由于胸导管引流液中细胞成份和所处状态与脾脏、扁桃体细胞不尽相同,我们对胸导管引流细胞制备IL-2的实验条件进行了探讨,实验结果表明:采用含5%人AB血清的完全培养液将细胞调整为5×10~6/ml的浓度,加PHA150μg/ml,于37℃5%CO_2饱和湿度培养48h,收集含IL-2上清,是制备粗制IL-2的最佳条件。利用胸导管引流细胞制备人天然IL-2,不仅活性高、产量大,而且较用人脾、扁桃体淋巴细胞制备IL-2手续简便。粗制IL-2初步纯化后制成的临床肌肉注射制剂,经临床志愿者试用后无不良反应。Since the cell coustituents and condition of FTDD are not different fromthose of the cells from spleen and tonsil in all respects, we studied the conditionsto produce IL--2 contained in supernatant. The results showed that 5×10~6/mlcells with 150 μg/ml PHA in the complete medium of 5% human AB serumwere cultured for 48h at 37°C and with 5% CO_2 in the air, and that was thebest condition to produce IL-2 contained in supernatant from FTDD. UsingFTDD to produce IL--2 not only has higher activities and higher production ofIL--2, but also is simpler than using the cells from the spleen or the tonsil.In the muscle injection of IL--2 purified from IL--2 contained in superna-tant. no side effects were observed in volunteers.
