手法治疗大鼠膝骨关节炎的超微结构研究

摘　　要：目的 :观察手法对膝骨关节炎的预防及治疗效果。方法 :以 Wistar大鼠为实验对象 ,通过结扎大鼠双侧股静脉造成双下肢瘀血 ,骨内压增加 ,膝关节软骨细胞缺乏营养而导致退变的方法制造动物模型 ,随机分成空白对照组 ,手法干预组 ,透明质酸钠阳性对照组。手法干预组及透明质酸钠阳性对照组均于术后 4周起治疗 ,各组于术后 8周、1 6周取材 ,取股骨外髁进行透射电镜观察。结果 :手法干预组和透明质酸钠组软骨细胞发生胞浆空泡化 ,细胞器数量减少 ,功能减退以及细胞核固缩甚至细胞坏死崩解等退变现象的程度较空白对照组轻 ,手法组与透明质酸钠组相当。结论 :手法对预防和治疗膝骨关节炎有效 ,其效果与透明质酸钠关节内注射基本相同。Objective:To observe the effect of knee joint osteoarthritis by manipulation through transmission electron microscope.Methods:Twenty four Wistar rats were ligated both femoral veins,then randomized three groups,the group without treatment,the manipulation group and the sodium hyaluronate injection group.The last two were treated 4 weeks after operation.The animals were sacrificed at 8 and 16 weeks postoperatively.The external femoral condyle was excised and prepared for the transmission electron microscopic study.Results:The degenerative changes of articular cartilage in the manipulation group and the sodium hyaluronate group,such as vacuolar degeneration of cytoplasm,decreased quantity and degraded function of orginele,pycnosis of nuclear and necrosis even disintegration of cell,are lesser than that in the group without treatmnet.Conclusion:Manipulation is effective to prevent and treat knee joint osteoarthritis well as intraarticular sodium hyaluronate injection.
