立克次体感染的PCR检测

摘　　要：作者应用ＰＣＲ技术对３０例疑似立克次体感染或不明热患者的血液标本和／或立克次体细胞分离培养物进行立克次体特异性ＤＮＡ检测。用Ｒｒ．１９０．７０ｐ、Ｒ．ｒ．１９０６０２ｎ，Ｒｒ．１７ＫＤ，ｓｔａ５８３种引物对分别进行ＤＮＡ扩增，共检出斑点热群立克次体特异性ＤＮＡ１１份，斑疹伤寒立克次体特异性ＤＮＡ４份，恙虫病立克次体特异性ＤＮＡ４份，其中有１０份血液标本在分离前用Ｒｐ．Ｃ．Ｓ．引物进行ＰＣＲ检测，检出立克次体属ＤＮＡ７份，分离培养后再取分离培养物和阳性血液标本同法用Ｒｒ．１９０．７０ｐ、Ｒ．ｒ．１９０６０２ｎ，Ｒｒ．１７ＫＤ，ｓｔａ５８３种引物对分别进行ＰＣＲ检测。Blood samples and/or isolated rickettsial cell cultures from 30 patients suspected of rickettsial infection or unknown fever were studies for rickettsial specific DNA by PCR with 3 pair primers R.r.190.70 p、R.r.190.602 n,R.r.17 KD,and sta58.As a result,R.SFG specific DNA were detected in 11 samples,R.typhus specific DNA were noticed in 4 samples,and R.tsutsugamushi specific DNA were found in another 4 samples.10 of these samples showed rickettsial family specific DNA by PCR with primers R.p.C.S before rickettsial isolation by cell culture and 7 of them were positive.All the cell cultures and the positive blood samples were detected again in the same way by PCR with 3 pair primers of R.r.190.70 p、R.r.190.602 n,R.r.17 KD,and sta58 and the results were correspondent to those before the isolation.
