人尿集落刺激因子的纯化及性质

摘  要:本文报道了从人尿中提取纯化获得一种CSF,它能促进小鼠骨髓有核细胞增殖分化,形成粒巨噬系细胞集落。提取纯化方法包括超滤浓缩、硫酸铵盐析、DEAE-纤维素柱层析、凝胶过滤及聚丙烯酰胺凝胶电泳等,所得CSF纯化约1500倍。经SDS-PAGE测定分子量为60,000道尔顿,用等电点聚焦技术测得pI为4.35,比活性为2.47×104U/mg蛋白质。人尿中存在集落抑制因子(CIF),该因子可经DEAE纤维素柱层析与CSF有效分离。经集落形成抑制试验证明,CIF对CSF刺激粒巨噬系祖细胞的增殖分化具有明显的抑制作用。CSF that stimulated mouse bone marrow cells to proliferate in vitro and form colonies of granulocyteand /or macrophage was purified about 1500-fold from normal human urine. Purification proceduresincluded concentration with ultrafiltration, ammonium sulfate precipitation, chromatographic separa-tion on DEAE-cellulose, and gel filtration. The molecular weight of the purified factor was estimatedto be about 60000 dalton by SDSPAGE, the p1 was estimated to be pH 4. 35 by isoelectric focusing,and the specific activity was found to be 2. 47× 104U/mg protein using mouse bone marrow cells,re-spectively. A urinary colony inhibiting factor (CIF)was separated from the CSF on the DEAE-cellu-lose column. This CIF suppressed the formation of granulocyte/ macrophage colonies of mouse bonemarrow cells when employed in conjunction with the purified urinary CSF.
