摘  要:目的:观察培养的新生小牛右心室心内膜内皮细胞(EEC)血管活性物质的分泌功能及缺氧对其分泌的影响。方法:EEC采用刮取法培养,用放免法测定培养液中血管活性肽的含量。结果:缺氧24h(PO2约为0.8kPa)使EEC的内皮素(ET-1)和心钠素(ANP)分泌显著增加(P<0.01),其分泌分别是常氧的1.9倍和1.6倍;缺氧也使EEC分泌内源性洋地黄因子(EDLF)显著增加(P<0.05),其分泌是常氧的1.7倍,但缺氧使EEC分泌血管紧张素(ATⅡ)降低18%(P<0.05)。结论:缺氧可引起EEC多种血管活性因子的分泌发生改变。Objective: To explore the effects of hypoxia on secretion of vascular activated peptide by cultured endocardial endothelial cells (EECs). Methods: After EECs were extracted from the right ventricule with scraping and cultured, the content of vascular activated peptides in the culture liquid was determined. Results: At the 24th h after hypoxia, the secretion of endothelin1 (ET1) and atrial natriuretic peptide (ANP) by EECs was significantly increased (P<0.01) to levels which were respectively 1.9 and 1.6 fold of those under normoxia. The secretion of endogenous digitalislike factor (EDLF) by EECs was also markedly increased (P<0.01) to a level which was 1.7 fold of that under normoxia but the secretion of angiotensin II (AT II) was decreased by 12%. Conclusion: Hypoxia results in changes of the secretion of various vascular activated factors by EECs.
