摘 要:目的:观察放烧复合伤小鼠骨髓基质细胞贴壁层的粘附能力及对GM-CFU(粒单系造血组细胞集落)形成的影响。方法:采用细胞粘附实验和细胞粘附阻断实验方法。检测5Gyγ线全身照射、15%体表面积Ⅲ度烧伤和放烧复合伤小鼠骨髓基质细胞粘附的功能。结果:单烧组伤后第3~7天显著高于正常组(P<0.05~0.01);单放组伤后第3~7天显著低于正常组(P<0.05~0.01);复合伤组伤后第3天显著低于正常组(P<0.01);正常组骨髓基质细胞贴壁层分别加入VCAM-1、Fn、Ln、ColⅣ等单抗或VCAM-1单抗分别联合Fn、Ln和ColⅣ等单抗处理后,GM-CFU数量均有不同程度的降低。Objective: To investigate the adherent capacity of mononuclear cells and GM CFU growth on stromal cell adherence layer in the bone marrow after combined radiation burn injury using in vitro culture. Methods: The bone marrow stromal cells extracted from mice inflicted with 5.0 Gy γ ray radiation injury, 15% TBSA full thickness skin burns and combined radiation burn injury respectively were cultured and detected with cell adhesion assay and cell adhesion blocking technique. Results: The adherent capacity of the mononuclear cells from the mice with simple burns was higher than that from the normal controls and significantly increased at the period from the 3rd to 7th d after injury as compared with those from the normal controls, mice with simple radiation injury and mice with combined radiation burn injury (P<0.05~0.01). Both of the last two were significantly decrease as compared with that from the normal controls (P<0.05~0.01). After the stromal adhesion layer in the bone marrow from the normal controls was added with such monoclonal antibodies (McAbs) as VCAM 1, Fn, Ln and Col IV respectively or VCAM 1 in combination with Fn, Ln and Col IV respectively, the number of CFU GM was decreased in various degrees. Conclusion: The decrease in adherent capacity of bone marrow hematopoietic indusive microenvironment might be one of the important reasons for hematopoietic disorder after combined radiation burn injury.
