小柴胡汤对荷瘤鼠S_(180)细胞生长和结构影响的研究

摘　　要：目的：通过体内实验，探讨小柴胡汤（ＸＣＨＴ）浸膏对荷瘤鼠Ｓ１８０的抑制作用。方法：采用透射电镜观察小柴胡汤对瘤细胞生长和结构的影响。结果：高、中和低浓度ＸＣＨＴ（１０００ｇ·Ｌ－１、５００ｇ·Ｌ－１、２５０ｇ·Ｌ－１）溶液治疗后，对荷瘤鼠Ｓ１８０细胞均有明显抑制作用，抑瘤率４０１９％～５８７３％，其中以高浓度组疗效最佳。同时可见经ＸＣＨＴ治疗后各组Ｓ１８０细胞均出现凋亡的特征，表现为细胞体积变小，核固缩，胞浆和核染色质凝集，密度增高，细胞间有大量凋亡小体，其中以低浓度组为甚，而高浓度组Ｓ１８０细胞则以坏死为主。结论：ＸＣＨＴ对荷瘤鼠Ｓ１８０有明显抑制作用，可诱导Ｓ１８０细胞凋亡及坏死，其中瘤细胞以哪种死亡方式为主。AIM and MITHODE: We observed the effect of Xiao Chai Hu Tang(XCHT) extract on tumor weight and ultrastructural change by electron microscopy to identify whether XCHT could induce apoptosis of S 180 in bearing tumor mice. RESULTS: After treatment with XCHT solution of high, middle or low-concentraction (1000 g/L,500 g/L,250 g/L),it could markedly inhibit S 180 growth (especially in the high concentraction group). Rate of inhibition ranged from 40.19% to 58 73%. Meanwhile, the S 180 in all of these groups (especially in the lower concentraction group) showed characteristic features of apoptosis which displayed by tumor cell decreased in size, karyopyknosis, cytoplasmic and nuclear chromatin condensation, density increased and a lot of apoptotic bodies among cell. Whereas the way of S 180 death in high-concentraction group was mainly degeneration and necrosis.CONCLUSION:The results suggested that XCHT had antitumor effect. It could induce S 180 cell death, including apoptosis and necrosis. The way of S 180 death was defferent depending mainly on dose of XCHT administered.
