5－Fu福氏佐剂包裹物抗肿瘤作用的实验研究

摘　　要：本文利用福氏佐剂包裹化疗药５－Ｆｕ，制成５－Ｆｕ－ＦＣＡ，对其体内外释放进行了研究，与５－Ｆｕ注射液比较体内、体外均有明显缓释作用。并观察了５－Ｆｕ－ＦＣＡ对艾氏腹水瘤、Ｓ１８０肉瘤（腹水性）及Ｈ２２小鼠肝癌瘤（腹水性）、荷瘤鼠的抗瘤作用，发现于接种瘤株后第２日一次性注射５－Ｆｕ－ＦＣＡ（４ｍｇ／只），小鼠存活期显著延长，且每组均有２只完全治愈；与盐水ＦＣＡ、５－Ｆｕ组比较有显著差别（ｐ＜０．００１）。该研究为提高５－Ｆｕ治疗效果提供了一个新途径。The release of 5-Fu-FCA,antitumor drug 5-Fu entrapped by FCA (Freund's Complete Adjuvant),was studied in vivo and in vitro.Compared with 5-Fu injection,5-Fu-FCA showed singnificant slow-release effect.The antitumor effects of 5-Fu-FCA on S180,EAC and H22 tumor bearing mice were observed.The results showed that after intraperitoneal injection of a single dose of 5-Fu-FCA (4mg/mouse)one day post-inoculation of tumor cells, the average survial times of S180,EAC and H22 tumor bearing mice treated with 5-Fu injection were 18.3±1.6.17.9 ±1.9and 19.1±3.6days,respectively, none of them were cured. While those of 5-Fu-FCA treated groups were 41.6±15.5,30.6±18.5 and 33.4 ± 21.6days, and 2/7,2/8,2/8 of the tumor bearing mice were cured respectively (VS 5-Fu treated groups, p<0.001).This study provides a new method for augmenting the antitumor effects of 5-Fu.
