摘  要:目的 探讨核酸疫苗在 HCV感染中的预防和治疗价值 .方法 将 HCV C区基因插入 p EF质粒 EF- 1α启动子下游 ,构建 p EF- HCV C重组质粒 .然后经 im及皮下注射免疫 BAL B/ c小鼠 ,免疫次数分别为 1,2 ,3,4次 ,EL ISA法检测免疫小鼠血清中抗 - HCV的水平 .结果 接种后各组均检测到抗 - HCV,但是抗体滴度较低 .在不同的免疫次数中 ,4次免疫组抗体水平最高 .结论 构建的 HCV DNA重组体可诱发 BAL B/ c小鼠产生体液免疫应答 ,加强免疫后可使血清中抗 - HCV明显增高 .AIM To study the protective and therapeutic value of DNA vaccine in HCV infection. METHODS The recombinant plasmid pEF-HCV was constructed by inserting the highly conserved HCV-core gene fragment into the eukaryotic expression vector pEF under the control of an EF-1α promoter, and then was injected into the quadriceps muscles or inoculated subcutaneously into right and left flank of BALB/c mice. Antibody to the HCV core protein in serum was detected by ELISA. RESULTS Antibody to HCV core protein was detected in serum of all the mice receiving injection of recombinant pEF-HCV-C, but the titers were low. The serum antibody level rose significantly after the booster injection, the antibody levels of mice with four times inoculation were the highest. CONCLUSION The results suggested that HCV core DNA-based immunization can induce HCV specific antibody, but the humoral immune response was weak at early stage, and the booster immunization can induce the rise of the antibody significantly.
