摘  要:用中华绒螯蟹(Eriocheirsinensis)未受精卵、受精卵、二细胞胚、囊胚、原肠胚为材料 ,提取总RNA ,分别用OligodT12GA、GC、AC3种锚定引物合成以上5种材料的cDNA第一链 ,然后以此cDNA为模板用30种随机引物进行PCR扩增 ,琼脂糖凝胶电泳分析。得到了5个早期发育的特异表达片段。结果表明 ,未受精卵、受精卵、二细胞胚之间的差异不大 ,都表现为母型调控 ,囊胚期有新mRNA的转录 ,开始进入合子型调控。The mRNA differential display technique was used to analyse the changes in gene expression that occurred in the process of early embryogenesis of Eriocheir sinensis. The total RNA was isolated from 5 kinds of material: unfertilized eggs, fertilized eggs, 2 cell eggs, blastula, gastrula. The first strand of cDNA was obtained by reversed transcription using three archored primer:Oligod T12AC, Oligod T12GA, Oligod T12GC. 30 arbitrary 10 mer primers were used to amplify the cDNA, and 5 special fragments, which related to the gene expression in early developmental embryo, were identified by agarose gel electrophoresis. The results demonstrated the differentiation among unfertilized eggs, fertilized eggs, 2 cell eggs were not very obvious, all of them were maternal regulation. New RNA was showed in the blastula, zygotic regulation came into being in this stage.
