检测血清弓形虫IgG抗体的滴金免疫测定法的建立及应用

摘　　要：目的 :建立检测血清中弓形虫IgG抗体的简便、快速的滴金免疫测定法。方法 :将弓形虫抗原包被于硝酸纤维素膜上 ,通过胶体金 -SPA直接显色 ,阳性者在膜上出现红色斑点。结果 :用该法与间接ELISA法平行对比检测经间接血凝法确定的 11份阳性血清和 30份阴性健康人血清 ,结果有良好的一致性 ,符合率为 90 .9%。结论 :滴金免疫测定法检测血清中弓形虫抗体简便、快速、敏感 ,无需特殊仪器设备 ,适于基层应用。Objective :To establish a simple,rapid and sensitive method for the detection of anti Toxoplasma gondii IgG in serum. Method A dot immunogold filtration assay (DIGFA) was developed by coating Toxoplasma gondii antigen on a nitrocellulose membrane.Tested serum sample was added and the membrane was visualized with SPA Colloidal gold.Results:The DIGFA technique was used in comparison with the indirect ELISA method to detect 11 positive and 30 negative serum samples confirmed by IHA method,and a good consistence was obtained,with a coincidence rate of 90.9%.Conclusions DIGFA is simple,sensitive and rapid for the detection of anti Toxoplasma gondii IgG and need no special instruments.
