摘  要:目的 探讨皮瓣微血管的酶组织化学染色效果。方法 应用酶组化法对人体皮瓣的微血管进行硷性磷酸酶染色。结果 组织用 10 %缓冲福尔马林 (甲醛 ) 4℃固定 6~ 12小时 ,冰冻切片 30~ 5 0 μm ,酶显示微血管的效果最好 ;皮瓣的微血管主要分布于真皮乳头层、网状层及皮下层 ;血管壁呈棕褐色 ,微动脉及毛细血管着色深 ,微静脉着色浅。结论 该方法不需灌注 ,对血管无扩张作用 ,无人为改变 。Objective To investigate the effects of enzyme-histochemical staining on microvessel of skin flap. Methods The histochemical staining of alkaline phosphatase was used to show microvessel of human body′s flap. Results The best result was obtained when tissue specimens were fixed in 10% buffer formalin at a temperature of 4℃ for 6 to 12 hours, vibratoned into 30~50μm microv-section and then stained with alkaline phosphatase; The microvessels were mainly distributed in the papillary, the dermal reticular and subcutaneous layers,the microvessel walls were brown stained, with dark color in microarteries and capillaries, and light color in microveins. Conclusion This method can be used to evaluate blood circulation quantitatively or semi-quantitatively in tissue or organs, through measuring the density of their microvascular bed without artificial errors, such as dilatation and rupture of blood vessels resulted from perfusion.
