摘 要:目的 :为探讨肾生髓的分子基础 ,阐述肾生髓的本质相关基因 ,我们观察了补肾生血药对衰老小鼠骨髓粒细胞 巨噬细胞集落刺激因子 (GM CSF)基因转录水平的影响。方法 :采用老年小鼠和D 半乳糖致衰老小鼠两种衰老肾虚动物模型 ,分别连续灌服补肾生血药 2个月和 1个月 ,应用半定量逆转录 聚合酶链式反应技术 (RT PCR)检测小鼠骨髓有核细胞GM CSF基因转录水平的变化。结果 :老年小鼠和D 半乳糖致衰老小鼠骨髓有核细胞GM CSF基因转录水平明显低于青年小鼠和正常对照小鼠 ,给药后则可明显得到提高。结论 :补肾生血药起到滋阴补肾、益髓生血作用的分子机理之一 ,是促进骨髓GM CSF基因的转录 ,GM CSF是肾生髓的分子基础之一 ,GM CSF基因是肾生髓的本质相关基因之一。Objective: In order to explore the molecular basis of the theory that “The kidney Produces the Marrow” and clarify the essential gene related to that,the effect of Bu Shen Sheng Xue Yao (BSSXY, Traditional Chinese Medicine, ripe for tonifying the kidney to promote hematopiesis) on the level of granulocyte macrophagocyte colony stimulating factor (GM CSF) gene transcription in marrow cells of aged mice was studied. Methods: After natural aged mice and D gal's aging mice were medicated with BSSXY for two months and a month respectively,the transcription level of GM CSF gene in marrow cells was measured by the techniques of semi quantitative reverse transcription and polymerase chain reaction (RT PCR). Results: The transcription level of GM CSF gene in the marrow cells of natural aged mice and D gal's aging mice were significantly lower than those of young and normal control. After medication with BSSXY, they were obviously improved. Conclusions: One of the mechanisms of BSSXY's effects is to promote the transcription of GM CSF gene. GM CSF is one of the molecular basis of the theory that “ The kidney Produces the Marrow”. GM CSF gene is one of the essential genes related to the theory above.
