补肾方药对去卵巢大鼠骨质疏松的骨组织ER表达的影响

摘　　要：目的 :探讨雌激素受体在骨质疏松发病中的作用及补肾方药对其调整作用。方法 :通过切除大鼠卵巢造成骨质疏松模型 ,补肾方药和尼尔雌醇分别灌胃治疗 3个月 ,观察补肾中药对骨质疏松的治疗作用。并且采用免疫组织化学 ,RT -PCR方法检测大鼠去卵巢前后骨组织中雌激素受体的变化 ,及补肾中药治疗对其的影响。结果 :卵巢切除后大鼠的血清雌二醇水平、腰椎中的雌激素受体表达水平及股骨干骺端雌激素受体mRNA表达均下降。补肾中药能够提高去卵巢大鼠的骨密度 ,改善其病理变化 ,提高大鼠血清E2水平 ,并且可以从RNA、蛋白质两种水平 ,提高骨组织中的ER的表达。结论 :补肾方药对去卵巢实验性骨质疏松大鼠有治疗作用 ,其作用机制不仅与其拟雌激素样作用有关 ,而且与其提高ERα在骨组织中的表达有关 。Objective:Explore the changes of the level of estrogen receptor in ovariectomized osteoporosis rats and the regulation of tonifying kidney recipes. Methods:The SD rats were ovariectomized and administrated tonifying kidney recipe and estrogen for 12 weeks. The ER protein was assessed by immunohistochemical and the expression of ER mRNA of bone was semi quantified by RT-PCR. Results: The bone mineral density, E\-2 and the expression of ER of the ovariectonized rats are decreased significantly and that of the rats herb treated did not decreased. Conclusion:Tonifying kidney recipe can treat the ovariectonized rat osteoporosis,it's partial mechanism effect was that it not only has solt estrogen like effect but also can up regulate the expression of ER not only in protein protein but also in mRNA level.
