摘  要:目的 观察许旺细胞源神经营养因子的超微结构定位 ,探索其在神经、皮肤和肌肉中的分布以及神经损伤前后量的变化。 方法  (1)胶体金标记免疫电镜法对预损伤 2周大鼠坐骨神经超薄切片进行许旺细胞源神经营养因子的亚细胞定位 ;(2 )免疫金银染色法对正常及损伤 2周后的坐骨神经、肌肉和皮肤进行许旺细胞源神经营养因子的标记并观察量的变化。 结果  (1)在许旺细胞胞浆中有明显的均匀圆形的致密金颗粒附着 ,但大量存在于内质网和高尔基体上 ;(2 )对照组、正常及失神经支配肌肉组为 (- ) ,正常神经组为 (+) ,正常皮肤组为 (++) ,变性神经组和失神经支配皮肤组为(+++)。 结论  (1)许旺细胞中 ,内质网可合成许旺细胞源神经营养因子 ,并且在高尔基体加工 ;(2 )神经和皮肤中存在有许旺细胞源神经营养因子 ,并且损伤后有量的明显增加 ,但在正常或失神经支配肌肉中没有许旺细胞源神经营养因子表达。正常皮肤中许旺细胞源神经营养因子含量高于正常神经 ,失神经支配皮肤中许旺细胞源神经营养因子含量相当于变性神经。Objective (1)To study the ultra structure localization of Schwann cell (SC) derived neurotrophic factor (SDNF) in Schwann cells(2)To investigate the distribution and quantitative changes in nerve、skin and muscle before injuries and after injuries Methods (1)Four SD rats were transected one side of sciatic nerve,the distal degenerative nerve were obtained after two weeks and made to ultra slicesAfter labeled by colloidal gold,the ultra slices were observed under transmitted electromicroscope(2)One side of sciatic nerve of ten SD rats were transected,after two weeks,both of sciatic nerves、gastrocnemius and dorsum pedis skins,no matter injured or uninjured,were obtained and made to frozen sectionsAfter treated with immunogold-silver stain (IGSS),the slices were observed under microscope Results (1)There were obvious high electro-density gold-granules in the cytoplasm of the SC,but the most on the endoplasmic reticulum and Golgi apparatus(2)There were silver staining in nerve and skin,but not in muscle Conclusions (1)SDNF can be synthesized in the endoplasmic reticulum and processed in the Golgi apparatus of SC(2)SDNF can be found in the normal and denerved skin,but not in the normal and denerved muscle
