猪细小病毒单克隆抗体杂交瘤细胞株的建立及初步应用

摘　　要：用经超滤浓缩、PEG 沉淀,氯化铯密度梯度离心纯化的猪细小病毒(PPV)免疫 BALB/C 小白鼠,取免疫鼠脾细胞与骨髓瘤细胞 SP_2/O在PEG 存在下融合,经 HAT 筛选、ELlSA 测定及3次克隆化后,获得3株(P-15—36—6,P-15—45—51,P—15—52—28)能持续稳定分泌抗 PPV 单克隆抗体的杂交瘤细胞株,3株杂交瘤细胞的染色体数均在95条以上,经分类测定,其免疫球蛋白均属 Ig G,且均为 IgG_1亚类。阻断试验证明单抗具有抗 PPV 的特异性。利用间接 ELISA 抑制试验进行了单抗在诊断上应用研究。BALB/C mice were immunized with porcine parvovirus which had been purified by ultrafiltering concentration,precipitation in PEG and ultracentrifugation in CsCl gra- dient.After HAT selection,cloning three times and ELISA test,three positive hybridoma cell lines(P-15-36-6,P-15-45-51,P-15-52-28)were established from the fusion of murine myeloma cells( SP_2/O)and spleen cells of BALB/C immunized with PPV.Their chromosome are more than 95 and their immunoglobulin class/subclass are all IgG_t.Ac- cording to bloking test,we recognized that the McAb secrected by the cell lines No. P-15-45-15 show a speciality to PPV.It has been investigated that the monoclonal antibody was applied in detection of PPV by the indirect ELISA inhibiting assay.
