低强度红激光局部照射对血管成形术后血管内皮功能的影响

摘　　要：目的　观察低强度红激光照射 (LPRLI)对动脉粥样硬化兔血管成形术后血管内皮修复和功能的影响。方法　新西兰白兔 40只 ,随机分为单纯球囊拉伤组和球囊拉伤并激光照射组各 2 0只 ,建立动脉粥样硬化兔血管损伤模型。激光照射组于球囊拉伤后应用激光球囊导管对损伤血管进行局部照射。对术后第 4周的两组动物 ,固定部位剪取拉伤组和激光照射组损伤的腹主动脉血管 ,采用离体血管功能实验方法 ,观察兔腹主动脉环的张力变化和损伤血管内皮细胞的修复情况。结果　经过LPRLI的兔损伤腹主动脉的内皮依赖性舒张功能明显强于单纯拉伤组血管。右髂动脉损伤段血管损伤面积 ,单纯拉伤组 (6 1 11± 7 12 )mm2 ,激光照射组 (96 71± 8 2 0 )mm2 ,后者的修复面积明显大于前者(P <0 0 1)。Objective To investigate the effects of low power red laser irradiation on endothelial function and repairment of regenerated endothelium after balloon angioplasty of rabbit abdominal aorta. Methods We established atherosclerotic arterial stenosis models successfully. 40 male New Zealand white rabbits were randomized into two groups: balloon injury group and balloon injury plus laser irradiation group (total irradiation density: 0.9 J/cm 2). After 4 weeks, the rabbits were killed, the abdominal aortas were removed and 2 mm vascular rings were inmersed in cold Krebs solution. The rings were progressively stretched for 2 hour until the optimal length tension relation was achieved as assessed by contraction to phenylephrine (Phe). To study vasodilation, the vessel was first contracted by a concentration of Phe inducing the half marximal response. Endothelium dependent relaxations to acetycholine (Ach)were studied in the rings from the lesion portion with LPRLI and the corresponding control rings. Relations to sodium nitroprusside (endothelium independent relaxation) were studied in a same way. All the rings pretreated with L Arg or L NMMA were incubated with Ach in order to find the relationship with NO. The 4 th week rabbit's iliac arteries subjected to balloon injury were determined using Evan's blue staining of the arterial surface. Photographs were enlarged and percentage reendothelialization was calculated using a digital planimeter as a percentage of the total areas. Results (1)Organ chamber experiment: Endothelium dependent responses of the rabbit abdominal artery were characterized in preliminary experiments. Acetylcholine (Ach), sodium nitroprusside (SNP), L Arg (L Arginine)+Ach, N G L monomethyl L arginine (L NMMA)+Ach caused concentration dependent relaxations in artery rings. After LPRLI, indothelium dependent relaxations to Ach and pretreated L Arg+Ach were significantly improved compared with relaxations in control rings at 4 th week (30 4�
