摘  要:目的 :探讨重组sICAM - 1对大鼠白细胞与肺微血管内皮细胞粘附的影响。方法 :采用原代培养的大鼠肺微血管内皮细胞及99Tm -HMPAO标记的白细胞 ,观察不同浓度的sICAM - 1、CA7(sICAM - 1的单抗 )以及sICAM - 1·CA7(sICAM - 1的二聚体 )对PMA刺激的大鼠白细胞与肺微血管内皮细胞粘附的影响。结果 :sICAM - 1与CA7即使在 10 0mg/L也不能抑制白细胞与肺微血管内皮细胞的粘附 ,而其二聚体在 2 0mg/L和 40mg/L即可抑制42 %和 5 0 %的细胞粘附 (P <0 0 5 )。结论 :sICAM - 1对细胞粘附的抑制作用很弱 ,可能与其单体形式有关。AIM: To explore the effect of recombined sICAM-1 on the adhesion of leukocyte to pulmonary microvascular endothelial cell. METHODS: Primary cultured rat pulmonary microvascular endothelial cells were used in this experiment. Leukocyte was labeled with 99 Tm-HMPAO.The effects of different dose of sICAM-1, CA7 and dimeric form of sICAM-1 (sICAM-1·CA7)were tested separately in cell adhesion .RESULTS: sICAM-1 and CA7 could not inhibit cell adhesion even at the concentration of 100 mg/L,while sICAM-1·CA7 at concentrations of 20 mg/L and 40 mg/L could inhibit 42% and 50% of cell adhesion respectively ( P< 0.05). CONCLUSION: sICAM-1 has poor inhibitory effect on cell adhesion, probably due to monomeric form.
