摘  要:目的:探讨谷氨酸致伤后大脑皮层神经完胞内游离钙的动态变化特征.方法:大鼠大脑皮层神经元原代分离培养方法和光电联合检测技术,培养8天的神经元用于实验.结果:谷氨酸致伤2h内,神经元胞内游离钙降低,60min、90min组比对照组明显降代,2h后胞内游离钙升高,以210min、480min组升高显著.结论:Ca2+稳态破坏参与谷氨酸致伤过程,不仅是Ca2+超载,早期Ca2+降低值得重视.Objective: To observe the effect of glutamate on the calcium homeostasis of cultured rat cortical neurons. Method:Single neuron culture technology and light-electron measurement system. The cultured neurons in 8 day were uscd the experiment.Result: The Ca2+ level was decrease in 2h after glutamate exposure The level was lower than that of control in 60min and 90min but higher in 210min and 480min. Conclusion:disturbance of calcium homeostasis is contributed to the neuron injure induced by glutarnate. The Ca2+ decrease in the early period has value in neuron injure. The cytosolic free calcium Ca2+ change processes were observed at the time points of 50, 90, 120, 150, 210 and 480 min after exposure to 300μmol/L glutamate for 5 min.
