中药复方降酶合剂对大鼠肝脏细胞因子调节的实验研究

摘　　要：目的研究中药复方制剂降酶合剂对Ｄ－氨基半乳糖所致动物急性肝损伤的保护作用，和对内毒素（ＬＰＳ）刺激后肝 枯否细胞（ＫＣ）分泌ＩＬ－６、ＩＬ－８和ＴＮＦ－а的调节作用。方法　复制大鼠Ｄ－氨基半乳精致急性肝损伤模型，制备ＳＤ大鼠 降酶合剂和联苯双酯的含药血清。以ＬＰＳ和含药血清刺激ＳＤ大鼠体外培养ＫＣ，采用ＥＬＩＳＡ夹心法检测ＩＬ－６、ＩＬ－８和 ＴＮＦ－ａ水平。结果Ｄ－氨基半乳糖所致急性肝损伤模型转氨酶和细胞因子分泌水平明显增高，中药降酶合剂可以降低转 氨酶和调节细胞因子分泌水平。培养细胞在加入ＬＰＳ刺激后，上清中ＩＬ－６、ＩＬ－８和ＴＮＦ－а浓度明显高于对照组（Ｐ＜０．０１）。 而在加入降酶合剂后，上清中的ＩＬ－６、ＩＬ－８和ＴＮＦ－а水平明显下降（Ｐ＜０．０１）。结论降酶合剂降低ＡＬＴ的作用可能与 其通过对机体免疫功能的调节，降低ＩＬ－６、ＩＬ－８和ＴＮＦ－ａ等细胞因子的产生有着密切的关系。Objective: To study the effect of Jiangmeiheji, a traditional Chinese medicine compound, on protecting liver from injury through rat acute hepatic injury model induced by D-galactosamine (D-Glan) and the adjusting effect of Jiangmeiheji on interleukin-6 (IL-6), interleukin-8 (ICS) and tumor necrosis factor a(TNF-а) secreted by Kupffer cells (KC) in response to endotoxin (LPS) stimulation. Methods The rat acute liver injury model induced by D-Glan was established and the sera of SD rats which had orally taken Jiangmeiheji and biphenyldimethylesterate respectively were prepared. The rats were stimulated by LPS and the prepared sera, and KC were isolated and cultured in vitro. The levels of IL-6, IL and TNF-a were measured by sandwich ELISA. Results The levels of ALT and cytokines in rat acute liver injury model induced by D-Glan were remarkably increased. Jiangmeiheji could reduce the ALT activity and adjust the cytokine levels. The contents ofIL-6, IL-8 and TNF-a in supernatant were higher than that in control groups (P <0.01) after the cultured KC were simulated by LPS; however, the levels of IL-6, IL-8 and TNF-a decreased obviously after Jiangmeiheji serum was added to the cultured KC (P <0.01). Conclusion Jiangmeiheji could reduce the production of such cytokines as IL-6, IL-8 and TNF-a by regulating the immune system to inhibit the activity of ALT.
