用Amp-Flp方法分析D1S80位点多态性

摘　　要：目的 :为了解广西壮族人群 D1S80位点群体遗传资料。方法 :使用扩增片段长度多态性 (Amp- Flp)和 PCR结合聚丙烯酰胺凝胶电脉及银染技术 ,对 30 0名广西地区壮族无关个体 D1S80位点多态性分析。结果 :观察到 2 3个等位基因 ,74个基因型 ,杂合度、非父排除率、个体识别力及多态信息含量分别为 0 .82 4、 0 .76 3、 0 .96 8、 0 .875。其基因型频率分布符合 Hardy- weinberg定律。对 5个家系相关个体分析符合孟德尔定律。结论 :该方法具有快速、简便、特异性强、灵敏度高特点。Objective:To obtain the distribution of allele and genotype frequencies of D1S80 VNTR Locus in Zhuang ethnic group in Guangxi.Methods:300 unrelated individuals in Zhuang were analyzed by PCR and PAGE following silver staining and amplified fragment length polymorphism.Results:23 alleles and 74 genotypes were observed in Zhuang population The observed Hetrozygosity(H),probability of exclusion(P E),discrimination power(DP) and polymorphism information content(PIC) were 0 824,0 763,0 968,0 875 respectively The observed genotype distribution was in accordance with Hardy weinberg equilibrium Conclusion:The method of Amp Flp is simple,rapid with high sensitivity and specificity
