猪小腔卵泡卵母细胞体外成熟研究

摘　　要：本实验通过与 2～ 6mm猪卵泡卵母细胞相对比 ,研究了猪 2mm以下卵泡卵母细胞的体外成熟。结果如下 :(1)由 2mm以下卵泡获得的小 (直径 10 0 2 9± 7 9μm)、中 (10 9 36± 3 71μm)和大 (118 90± 2 4 6 μm) 3类卵母细胞培养 4 8h的成熟率分别为 0、2 2 31%和 4 5 4 5%。 (2 ) 2mm以下卵泡卵母细胞的成熟率 (52 6 3% )显著低于 2～ 6mm卵泡卵母细胞的成熟率 (80 39% )。(3)以含PFF的培养液培养 2mm以下卵泡卵母细胞其成熟率 (53 54% )显著高于不含PFF的对照组 (2 2 77% )。 (4 )在培养的前 2 4h加入PMSG、后 2 4h去除PMSG ,其成熟率 (4 1 86 % )与含PMSG的培养液连续培养 4 8h的成熟率 (50 0 % )无显著差异 ,而显著高于前 2 4h不加PMSG ,后2 4h加入PMSG培养的成熟率 (5 4 1% ) ;也显著高于以下不含PMSG的培养液连续培养 4 8h的成熟率 (3 3% )。The in vitro maturation of the porcine oocytes from the <2 mm follicles were studied in comparison with those from the 2 5 mm follicles.The results are as follows:(1) The maturation rate of small (100.29±7 9 μm in diameter),middle (109 36±3 71 μm) and large(118 90±2 46 μm) oocytes from <2mm follicles was 0,22.31% and 45.45%,respectively,after cultured in vitro for 48 h.(2)The maturation rate (53.54%)of oocytes from <2 mm follicles cultured in the media with PFF was higher significantly (P<0 05) than that (22.77%) of the oocytes cultured without PFF.(3) The maturation rate (41.8%) of oocytes cultured with PMSG for the first 24 h but without PMSG for the second 24 h did not differ significantly from that (50 0%) of the oocytes cultured with PMSG for 48 h.This rate was higher significantly (P<0 05) than that (5 41%) of the oocytes cultured without PMSG for the first 24 h but with PMSG in the second 24 h,and was higher significantly(P<0 05) than that(3.3%) of the oocytes cultured without PMSG for 48 h.
