摘  要:目的 :用高效液相色谱 -蒸发光散射检测法 (HPLC -ELSD)直接测定银杏叶中银杏萜类内酯A、B、C及白果内酯的含量。方法 :银杏叶 10 %甲醇提取液通过聚酰胺及硅胶小柱联用净化制成供试品溶液 ,经C1 8色谱柱 ,以甲醇 -水为流动相 ,梯度洗脱 ,以蒸发光检测器检测。结果 :银杏叶供试品溶液的预处理是获得准确测定结果的关键 ,多点校正 ,自然对数换算以弥补ELSD线性应答差的不足。结论 :测定不同产地、不同树龄、不同采收季节的银杏叶中萜类内酯含量 ,对从源头抓好原料质量有积极意义。Objective:Ginkgolide A,B,C and bilobalide in Ginkgo biloba leaves are determined simultaneously by RP-HPLC-ELSD.Method:10% methanolic extracts of the leaves are cleaned-up by solid phase extraction via polyamide cartridge and silica gel cartridge successively and the sample solutions are prepared.RP-HPLC analysis is carried out through C 18 column,methanol-water as mobile phase,eluted in gradient mode,detected by evaporated light scattering detector.Result:The key step for getting precise data is pre-treatment of the leaves extract through normal phase SPE cartridge;The poor linear response of ELSD can be compensated by multilevel calibration and logarithm calculation.Conclusion:The Ginkgo biloba leaves obtained from different area,different growth years and collection seasons are determined.It is significant for quality control of the herbal medicine initiated from the raw material.
