摘 要:目的 :探讨脐血移植重建同种异基因纯系小鼠免疫造血系统的可行性 ,建立一种新的研究脐血移植的方法。方法 :孕 2 0d的C57BL/6小鼠 (H 2b)经剖腹产取出胎盘和胎鼠 ,分离出脐血单个核细胞(MNC) ,1× 10 6 个MNC经尾静脉注入致死量照射的BALB/c小鼠。 1月后 ,用抗H 2Db单克隆抗体检测嵌合体的存在。用混合淋巴细胞培养和皮肤移植检测特异性的免疫耐受。结果 :未移植的对照组 3周内全部死亡。移植组 75%存活并形成稳定的不全嵌合体。体外混合淋巴细胞培养和体内皮肤移植发现受鼠对供者的组织和细胞产生了特异的免疫耐受。结论 :小鼠脐血有足够的造血干细胞 ,能重建同种异基因小鼠的免疫造血系统而不引起明显移植物抗宿主病 (GVHD) 。Objective:To investigate the potential possibility to reconstitute the immunohematopietic system in adult mice recipients across major histocompatibility complex by near term cord blood and construct a new kind of method on the study of cord blood transplantation.Methods:1×10 6 fetal blood cells from 20 day old C57BL/6 pups(H 2b)delivered by cesarean were injected into the lethally irradiated (8.5Gy) BALB/c(H 2d).After one month,anti H 2Db antibody was used to investigate donor chimera in recipients.In vitro mixed lymphocyte response (MLR) and skin transplantation were used to investigate specific tolerance.Results:All of the control mice receiving lethal radiation died within three weeks after radiation.75% recipients survived without any clinical signs of graft versus host disease(GVHD) within the observation period of two months.Mixed chimera was demonstrated by using anti H 2Db antibody.The inability to reject the donor skin and the preservation of the capability to reject the third party skin indicated immune tolerance was established in the recipients.Immune tolerance was also confirmed by in vitro mixed lymphocyte response.Conclusion:Fetal blood contains enough hematopietic stem cells to reconstitute the immunohematopietic system in adult mice recipients across major histocompatibility complex without causing GVHD.This model may be suitable to address many questions raised in clinical cord blood transplantation.
