摘 要:采用递增皮下注射 (SC)吗啡 5天后 ,经纳络酮催瘾建立吗啡戒断模型 ,由眼内眦取血测定RBC变形能力 (ED)、血液粘度 (VIS)。结果表明 ,吗啡戒断大鼠ED较正常对照组明显下降、VIS明显升高 ,褪黑激素 (Melatonin ,MT)明显抑制吗啡戒断大鼠ED下降和VIS的升高。另外向正常大鼠血中直接加入不同剂量的吗啡、去甲肾上腺素 (NE)、环磷酸腺苷 (cAMP)也可使ED明显减小 ,而加入MT则可使ED明显增加 ,VIS下降 ,并呈一定的剂量效应关系。由此可见MT对吗啡戒断大鼠ED和VIS均有改善作用 ,这可能是通过MT对RBC的直接保护作用 ,也可能是抑制由戒断引起交感神经系统和G蛋白—cAMP系统亢进而发生的。A model of morphine dependent rats was established by gradually increasing the doses of morphine via subcutaneous injection(from 5mg·kg -1 to 50mg·kg -1 , 5days). Erythrocyte deformability(ED) and blood riscosity (VIS) of blood of rats were detected. Results showed that ED of morphine withdrawal rats was lower than that of the normal, and VIS was higher. Melatonin (MT) could inhibit the decreasing of ED and the increasing of VIS in morphine withdrawal rats. Moreover, if different doses of morphine, norephinephrine and cAMP were directly added into blood of normal rats, it could reduce the ED obviously, and could aggravate VIS also,but MT could improve all of those changes in a dose dependent manner. These results show that the improvement effect of MT on ED and VIS in morphine withdrawal rats could be results in by protecting RBC directly, or inhibiting the hyperfunction of the sympathetic system and protein G\|cAMP system.
