摘  要:目的 :观察在长期低浓度酒精处理后及撤除酒精给药的过程中 ,大鼠下丘脑及伏核中 β 内啡肽含量的变化 ,探讨酒精对 β 内啡肽系统的影响及其在酒精依赖过程中的作用。 方法 :以 6 % (v/v)的酒精作为大鼠饮水的唯一来源 ,分别取大鼠在饮酒和撤除酒精不同时程的下丘脑及伏核标本 ,用放免法测定每克组织中 β 内啡肽的含量。同时用顶空气相色谱法测定大鼠在饮酒过程中不同时段血中的酒精浓度。结果 :下丘脑及伏核的 β 内啡肽含量在长期 (4周 )饮用酒精后无明显变化 ,在撤除酒精后的 2~ 4天明显升高 ,1周后恢复正常。动物血中酒精浓度在摄入酒精的不同时段无明显差异。结论 :酒精影响下丘脑及伏核中 β 内啡肽的含量是低浓度反复多次刺激的结果 ,撤药后伏核内 β 内啡肽的高含量是动物产生依赖的重要原因。Objective:To investigate the effect of ethanol on the beta-endorphin system and their rat in ethanol dependence, the beta-endorphin content in the nucleus accumbens (Nacc) and hypothalamus of the rats during chronic ethanol expoured withdrawal was measured dependence. Methods:SD rats received drinking water containing 6%(v/v) ethanol for 28 days and withdrawal of this water on the 29th day. The Nacc and hypothalamus of the rats in different groups were obtained and their beta-endorphin content were also measured by radioimmunoassay (RIA). The ethanol concentration in the blood of the rats were also measured by headspace gas chromatography. Results:The beta-endorphin content in Nacc and hypothalamus had no change during the rats′ exposure to the ethanol, but obviously increased in the 2nd and 4th days after ethanol withdrawal. Conclusion:The alteration of the beta-endoephin content in Nacc and hypothalamus may result from the chronic and repeated exposure to low concentration ethanol. The high beta-endorphin content in the Nacc during the ethanol withdrawn may be essential to the athanol dependence in these animals.
