摘 要:为研究一氧化氮 /内皮素 (NO/ ET)失衡与肝缺血再灌注 (I/ R)损伤的关系以及磷脂酶 A2 抑制剂消炎痛(INDO)对 NO/ ET的调节作用 ,采用肝 I/ R模型 ,比较 I/ R组和 INDO组 NO/ ET的变化情况及其与肝 I/ R损伤的关系。再灌注急性期血 NO代谢产物 (NO2 - / NO3- )降低、ET升高致 NO/ ET比值降低 ,血 TNF- α、肝丙二醛 (MDA)及酶的漏出增加 ,肝 ATP含量降低 ,肝损伤加重 ;INDO可阻止再灌注后 NO2 - / NO3-下降、改善 NO/ ET平衡、减轻肝损伤。认为肝 I/ R损伤与 NO/ ET失衡有关 ,INDO对肝 I/ R急性期的保护作用与其对 NO/To study the relationship between the disturbance of nitric oxide/endothelin (NO/ET) and the hepatic ischemia/reperfusion (I/R) injury as well as the regulation of NO/ET by the phospholipase A 2 inhibitor indomethacin (INDO), the changes of NO/ET and their relationship with the hepatic I/R injury were compared between the I/R group and the INDO group in a hepatic I/R model. In the acute phase of hepatic reperfusion, the ratio of NO/ET was reduced, which was contributed to the significant reduction of NO 2 -/NO 3 - (the metabolic product of NO) but significant elevation of ET in the blood. TNF α in the blood, hepatic MDA and the leakage of enzymes were all increased but the hepatic ATP was reduced, the hepatic damage was deteriorated. INDO could improve the balance relationship of NO/ET by preventing the reduction of NO 2 -/NO 3 - and alleviate the injury of the liver. It was considered that the hepatic I/R injury might be concerned with the disturbance of NO/ET, the protection of INDO against hepatic I/R in the acute phase might be relative to its regulation on NO/ET.
