摘  要:目的 制备安全有效的A群脑膜炎球菌多糖结合疫苗。方法 通过碳二亚胺(EDAC)将A群脑膜 炎球菌多糖-AH衍生物与破伤风类毒素(TT)蛋白共价结合,制备A群脑膜炎球菌多糖结合疫苗。结果 所结合 成的多糖-蛋白结合物,具有A群脑膜炎球菌多糖和破伤风类毒素抗原特异性。单独用多糖免疫小鼠未能诱导出 高水平的血清Ps抗体,而用结合物免疫小鼠却诱导出高水平的血清Ps抗体及TT抗体,并有免疫记忆性。结论 为进一步临床评价该疫苗的人群免疫效果提供了实验基础。Objective To prepare a safe and effective meningococcal group A polysaccharide (Ps) conjugate vaccine. Methods The conjugate vaccine was prepared by the carbodimide-mediated covalent con pling of meningococcal group A polysaccharide-AH derivative to tetanus toxoid(TT) BALB/c mice were inject. ed with 10 μg of polysaccharide or conjugate vaccine,and the Ps and TT antibodies in the sera of them were detected by ELISA. Results The conjugate vaccine showed both Ps and TI' antigenicities. No serum Ps anti body at a high level was induced by polysaccharide vaccine,however, both serum Ps and TT antibodies at high levels were induced by the conjugate vaccine. The conjugate vaccine showed also immunological memory. Con clusion The study laid a foundation of further evaluation of the immunological effect of the conjugate vaccine in population.
