摘  要:以高粱细胞质雄性不育系A2 V4 (A)及其保持系V4 (B)的总DNA为模板 ,对 184个随机引物进行筛选 ,找到6个其RAPD扩增产物在A/B间存在稳定差异的引物 .将该 6个引物同时扩增A/B的总DNA、线粒体DNA(mtDNA)及叶绿体DNA(cpDNA) .以总DNA为模板时得到 12个扩增片段 ,以mtDNA为模板时得到 4个 ,以cpDNA为模板时得到 11个 .结果分析表明 ,在这些扩增片段中 ,有 7个仅仅出现在以胞质DNA为模板的扩增中 ,有 5个在以总DNA和胞质DNA为模板时同时出现 ,即认为这 12个片段来自胞质DNA .另有 7个片段 ,在以胞质DNA为模板时未出现 ,而是仅仅出现在以总DNA为模板的扩增中 ,认为是来自核DNA .来自核DNA的 7个扩增片段中 ,有 5个来自保持系 ,有 2个来自不育系 ,这表明 ,不育系与保持系在核DNA上存在差异 .对A/B核DNA在CMS中的重要性及研究对策进行了讨论 .图 2表 4参The total DNA from cytoplasmic male sterile line A 2V 4 (A) and its maintainer V 4 (B) in sorghum were screened primarily by RAPD using 184 random sequence 10 mer primers, and from which 6 primers, amplifying reproducibly polymorphic bands between A/B at each time, were identified. Then the 6 primers were used further to amplify their total DNA, mitochondrial DNA (mtDNA) and chloroplast DNA (cpDNA), respectively. And 12, 4 and 11 polymorphic fragments, which were different between A and B, were produced when total DNA, mtDNA and cpDNA were used as template,respectively. Excepting the 7 fragments appeared only in the of mtDNA and cpDNA, 5 among the 12 fragments occurred in total DNA also amplified in the amplification of mtDNA and/or cpDNA. And these 12 fragments were thought amplified from cytoplasmic DNA because of the total DNA including nuclear DNA, mtDNA and cpDNA. In contrast, another 7 fragments, which appeared only in the amplification of total DNA and never in that of cytoplasmic DNA, were thought amplified from nuclear DNA. Among these 7 fragments, there are 5 occurred in maintainer V 4 and 2 in CMS line A 2V 4, respectively. These results indicate it is different in nuclear DNA between CMS line and its maintainer. The importance of nuclear DNA on CMS and the significance of this researching strategy were discussed. Fig 2, Tab 4, Ref 19
