污染寄主卵面的斜纹夜蛾核多角体病毒PCR检测及其消除

摘　　要：ＰＣＲ法检测斜纹夜蛾核多角体病毒(Sｐｏｄｏｐｔｅｒａ　ｌｉｔｕｒａ　ｎｕｃｌｅｏｐｏｌｙｈｅｄｒｏｖｉｒｕｓ，ＳｐｌｔＮＰＶ）ＤＮＡ，其检测的最低量为 ５个病毒ＤＮＡ。经碱解处理并用 １０ Ｕ的 Ｄｎａｓｅ可以除去残留在每个卵表上 ９×１０４个多角体中所有的病毒 ＤＮＡ。利用这种方法可以除去污染昆虫卵表杆状病毒，为以后检测卵内是否存在 ＳｐｌｔＮＰＶ ＤＮＡ及 ＳｐｌｔＮＰＶ的流行病学研究奠定基础。PCR technology was employed to detect Spodoptera litura nucleopolyhedrovirus (SpltNPV) DNA sequences from viral polyhedron inclusion bodies (PIBs). The level of sensitivity by the technique was as low as 5 genome copies. The viral DNA on the egg surface coniaminated by polyhedra could be eliminated by treating with ten units of Dnase. This method may provide a better under- standing of SpltNPV epizootiology as well as the detection of the SpltNPV inside the egg or on the egg surface.
