摘  要:以湖北白猪卵母细胞为核移植受体,杜洛克胚胎卵裂球为核移植供体,进行了猪胚胎细胞核移植的研究,建立了核移植的程序。卵母细胞的电激活实验表明猪卵母细胞的激活以1.25 kV/cm、40 μs脉冲为最佳; 2-细胞期胚胎电融合实验表明脉冲时间 40 μs,1.5~20 kV/cm脉冲的融合效果较好。卵母细胞体外成熟时间对核移植效率有显著影响,42~48 h成熟卵做受体核移植效果较好。61枚核移植的重组胚胎移植给5头受体,1头维持妊娠至足月,顺利产下5只核移植猪。文中对核移植效率的影响因素和猪核移植的前景进行了分析。The nuclear transplantation was performed using oocytes of Hubei White pig as recipients and embryonic cells of Duroc pig as donors. An optimum program of nuclear transfer had been established for the pig. The best result of oocyte activation was gained at l pulse with strength l.25 kV/cm and lasting 40 μs, and the good resuits ofelectrofusion of 2-cell stage embryos were gained at l pulse with strength 1.5~2.0 kV/cm and lasting 40 μs. Aging of oocytes in vitro could significantly affect nuclear transfer efficiency, the optimum age of oocytes were 42~48 h. There were five foster pigs accepted 6l recombined embryos, only one of them developed to term, and five nuclear transfer piglets were born. The authors analyzed the factors affecting nuclear transfer efficiency and the prospects of nuclear transplantation of pig.
