摘 要:目的 :研究异种胶原膜 (戊二醛交联乙醇浸泡的Ⅲ型猪皮胶原生物膜 )作为兔鼓膜移植材料的生物相容性。方法 :1 光镜和透射电镜下观察兔耳正常鼓膜和生物膜的组织形态学结构 ;2 测定兔体液免疫抗体含量 ;3 测定生物膜的抗原性 ;4 以大耳白兔为受体 ,将生物膜植入背部肌肉 ,观察乙醇浸泡前后移植材料的生物相容性差别 ;5 切除动物鼓膜 ,移植生物膜鼓膜 ,于术后 1 0、2 0、40和 60d观察鼓膜愈合状态。结果 :1 证实兔鼓膜的组织学和超微结构与人耳鼓膜相似 ;2 兔体液免疫学抗体含量与人类的基本一致 ;3 生物膜的胶原蛋白抗原性低 ,其氨基酸符合胶原蛋白标准 ;4 生物膜具良好的网状结构 ;5 乙醇浸泡前后组织病理学反应差异有显著性 (P <0 0 5) ;6 移植组 1 9耳成功 ,空白对照组无 1耳愈合。结论 :异种胶原膜具有良好的生物相容性 ,是有实用价值的人类鼓膜移植材料。Objective:To probe into the biocompatibility of exogenous collagen (Type Ⅲ pig dermal collagen membrane cross-linked with glutaraldehy fixed in alcohol) as a rabbit's tympanic membrane graft.Methods:1.The histological and ultra-structural morphology of the rabbits tympanic membranae were observed under the light microscope and the transmission electron microscope. 2.The antigenicity antibody quantities of the serum in rabbits were measured.3.Biochemical and immunological assayed the graft.4.The graft was implanted into the deep layer of the back muscles of the rabbits to observe the biocompatibility before and after transplantation.5.The rabbits were removed the membranae tympani and transplanted the exogenous collagens;and they were observed the healing condition at the 10th,20th,40th and 60th day. Results:1.The histological and ultrastructural morphology of the rabbit tympanic membrane was similar to that of the human tympanic membrane.2.The quantity of antibody was similar to that of the human.3.A low antigenicity of the collagen in biomembrane was found and aminoacid analysis was in accord with the criteria for collagen protein.4.The collagen fibers of the biomembrane were of a good reticular devoid of other cells and blood vessels.5.The significant statistical differences in histopathology of the pre-and post-transplantation biocompatibility of the graft were observed(P<0 05).6.Successful transplantation was found in 19 of the 20 ears in study group,but none in the control group.Conclusion:Exogenous collagen has good biocompatibility and is a kind of valuable material for human membrane tympanic transplantation.
