摘 要:蚕丝丝素蛋白膜是一种具有弱碱性和弱酸性的两性荷电膜。在丝素膜-水溶液体系中,水溶液pH值变化会影响丝素膜荷电状况,进而影响到溶质的渗透速度和渗透量。5-氟脲嘧啶(5-FU)在外部溶液 pH<7时透过丝素膜的速度基本上维持常量,当外部溶液pH>8上下5-FU的渗透速度明显下降,表明在pH8上下5-FU的渗透状况呈开关状态。Permeation of 5-Fluorouracil through silk fibroin protein membrane was investigated. The membrane was prepared from Cocoon. Permeability coefficient of 5-FU was measure in the pH ranges from 3.0 to 9.0. The silk fibroin protein membrane was an amphoteric ion exchange membrane composed of both weak acid and weak base groups. The isoelectric point of this membrane was about pH 4.5. The measured results showed the permeability coefficient of 5-FU decreased about pH 8. It was due to 5-FU changed into anion 5-FU at alkaline solution. And the membrane was as cation exchanger membrane above pH 4.5. 5-FU anion was excluded by negatively charge group in the membrane. The permeation results of 5-FU were analyzed on the basis of TMS theory. According to theoretical analysis, the ion mobility of anion 5-FU in the membrane had distinctive changes at different pH, and apparent effective charge density in the membrane varied with pH changes. The permeate rate of 5-FU increased when pH change into 7, and decreased when pH return to 9, respectively.
