摘  要:目的 脐血移植是一种有前途的治疗手段。但在脐血移植中仍存在着单份脐血中干 /祖细胞的绝对数量少的问题。研究脐血细胞在体内生长的微环境 ,有助于最大限度的提高脐血细胞的扩增繁殖能力。细胞因子是脐血细胞造血微环境的重要组成部分 ,白细胞介素 6 (IL 6 )就是其中关键的一种。探讨脐血中IL 6的来源。方法 分别应用免疫组织化学染色和原位杂交的方法对 10例健康产妇足月分娩的胎盘组织及脐带和 7例人工流产的绒毛标本进行了对比研究。标本染色后 ,光学显微镜下观察 :细胞核呈紫红色 ,细胞浆有蓝色标记物的细胞为阳性细胞 ,细胞浆无蓝色标记物的细胞为阴性细胞。结果 足月胎盘滋养层细胞有IL 6的表达 ,而在脐静脉内皮细胞和绒毛标本均未见阳性表达。结论 脐血中IL 6主要是由胎盘滋养层细胞分泌 。Objective Cord blood transplantation is of prospect to treat hematopoietic diseases, immune deficiency syndrome and inherited metabolism disorders. However, there are still some problems unsolved in carrying this procedure, such as the amount of stem cells in one cord blood sample sometimes is insufficient for successful transplant. Cytokines, especially interleukin 6 (IL 6) can help enhance the expansion and reproduction of stem cells. Higher concentration of IL 6 has been found in cord blood compared to peripheral blood in adults. This study is mainly to investigate the possible cell source of this higher amount of IL 6. Methods Ten samples of term placenta tissue and umbilical cords were collected from ten health births, and other seven samples of villous were from seven artificial abortion cases. Frozen sections were made and the techniques of immunohistochemical staining and in situ hybridization were used for analysis. Mouse antibody to IL 6 antigen as primary antibody and biotinylated goat anti mouse antibody as secondary antibody were chosen in immunohistochemical staining. Negative control was designed to be incubated without primary antibody. For in situ hybridization IL 6 probes were. Labeled first were then hybridized with the post managed samples. Sample for contrast was designed to hybridize without labeled probes. Samples were observed under the microscope.Results Experiments indicated that the term placental trophoblastic cells had strong positive reactions while the samples of umbilical cord and villous showed negative reactions.Conclusion Trophoblastic cells in the placental tissue were the main production source of IL 6. The secreting function of trophoblastic cells might become mature with the growing of the embryo.
