血管生成因子在子宫内膜异位症中的作用

摘　　要：目的 :探讨血小板衍生内皮细胞生长因子 (PDGF)对体外培养的子宫内膜异位症患者子宫内膜腺上皮细胞的影响 ,及血管内皮生长因子 (VEGF)在内异症发生中的作用。方法 :对 11例子宫内膜异位症患者的子宫内膜组织进行体外培养、纯化、鉴定。将上皮细胞接种于 96孔板中 ,用加有不同浓度的PDGF处理。用MTT法观察PDGF对腺上皮细胞的作用并用计算机灰度分析仪分析VEGF的免疫组化结果。结果 :PDGF对腺上皮细胞有明显的促增殖作用 ,10ng ml时最明显 (P <0 .0 5) ,在不经过和经过PDGF刺激的腺上皮细胞中都有VEGF表达 ,但刺激后VEGF的表达明显增强 (P <0 .0 5)。结论 :PDGF和VEGF作为血管生成因子 。Objective:To explore the effect of platelet-derived growth factor (PDGF) on cultured eutopic endometrial epithelial cells and the effect of vescular endothelial growth factor (VEGF) in endometriosis.Methods:Eutopic endometrial cells of 11 women with endometriosis were cultured,purified and identified.Collected epithelial cells were treated with various concentrations of PDGF in 96 well plate.The effect was observed through MTT method.The immunohistochemistry results of VEGF were analysed by computer analyser.Results:PDGF was found to stimulate proliferation of epithelial cells and was most notable with 10ng/ml.VEGF can be detected in epithelial cells cultured with or without PDGF,but expression of VEGF in epithelial cells stimulated by PDGF is higher than that of control(P<0.05). Conclusions:As positive angiogenesis growth factors,PDGF and VEGF participate in pathogenesis of endometriosis by means of autocrine and paracrine.
