摘  要:为脐血造血干细胞 (CBHSC)的冻存建立简便有效的方法 ,采用 5 %二甲基亚砜 (DMSO)加 6 %羟乙基淀粉 (HES)为冷冻保护剂 ,不用程控降温装置 ,直接置CBHSC于 -80℃冰箱中降温 ,次日移至液氮液中保存 1周、1个月、3个月、6个月。结果冻存 6个月后 ,单个核细胞 (MNC)、粒 -单系造血祖细胞 (CFU -GM )和CD+34 细胞的回收率及MNC的台盼蓝拒染率分别为 90 .5± 1.7% ,87.6± 19.5 % ,91.9± 2 .9% ,75 .8± 1.8%。认为这一简便的方法能很好地保存CBHSC的造血潜能 ,因此它能为脐血移植冻存CBHSC。This article aims to establish simple and effective methods for the cryopreservation of cord blood hematopoietic stem cells (CBHSC). CBHSC was placed directly in a -80℃ electrical freezer with 5% dimethysulfoxide (DMSO) and 6% hydroxyethyl starch (HES) without a programme-controlled cooling apparatus. At the next day, these CBHSC were transferred into liquid nitrogen and stored for 1 week to 6 months. After the cryopreservation of cord blood cells for 6 months, the rates of recovery for mononuclear cells (MNC), for the granulocyte macrophage colony forming units (CFU-GM) and CD + 34 cells were (90.5±1.7)%, (87.6±19.5)%, (91.9±2.9)%, respectively, and the rate of trypan-blue chromophobewas (75.8±1.8)%. From this study it can be concluded that this simple method can effectivelyy maintain the proliferative capacity of CBHSC. Furthermore this method can be used to cryopreserve CBHSC for the CBHSC transplantation.
