连黛片对大鼠溃疡性胃癌相关癌基因蛋白表达的影响

摘　　要：目的：观察连黛片对大鼠溃疡性胃癌相关癌基因蛋白表达的影响，探讨连黛片防治胃癌的机理。方法：采用ＳＰ免疫组化法检测溃疡性胃癌模型组、连黛片治疗组大鼠胃癌组织以及溃疡对照组大鼠胃组织中ｃ－ｅｒｂＢ２、 ｐ２１ｒａｓ和 ｐ５３三种癌基因蛋白的表达情况。结果：所检 ６例模型组胃癌组织的 ｐ２１ｒａｓ， ｃ—ｅｒｄＢ２蛋白均呈阳性表达，而连黛片防治组２例胃癌组织的ｐ２１ｒａｓ，ｃ—ｅｒｂＢ２则呈阴性表达，两组胃癌组织的ｐ５３蛋白也呈阴性表达。另外所检５例溃疡对照组大鼠胃组织的上述三种癌基因蛋白，均呈阴性表达。结论：ｒａｓ、ｃ－ｅｒｂＢ２原癌基因的激活可能是Ｎ－甲基－Ｎ’－硝基－亚硝基胍（ＭＮＮＧ）诱发溃疡大鼠腺胃癌的重要原因，而连黛片则可能通过对抗 ｒａｓ、ｃ－ｅｒｂＢ２原癌基因的激活而抑制ｃ－ｅｒｂＢ２、 ｐ２１ｒａｓ蛋白的表达。Objective: To observe the effects of Liandai Tablet (LT) on relevant oncogenic protein expression of ulcerative gastric cancer in rats and to probe its anticancer mechanism. Methods: Three kinds of oncogenes, c - erbB2, p21ras and p53 in gastric tissues obtained from the modal group, treatment group and ulcer control group were detected by immunohistochemical method. Results: Protein expression of p21ras, c- erB2 in six samples from the model group was positive and that in two samples from the treatment group was negative' p53 protein expression in the above two groups was negative. c- erbB2, p21ras and p53 protein expression were negative in the ulcer control group. Conclusion: The activation of ras, c-erbB2 genes probably play an important role in the occurrence of rat MNNG-induced gastric cancer, and the mechanism of LT inhibiting the protein ex pression of c- erbB2, p21ras may be by counteracting the activation of ras, c- erbB2 genes.
