摘　　要：建立犬血浆中新型抗炎镇痛剂 SFZ-47[3 H -1 ,2 -二氢 -2 -( 4 -甲基苯胺基 )甲基 -1 -吡咯里嗪酮 ]浓度测定的 HPL C法 .选用 Kromasil C18色谱柱 ,流动相为甲醇 -磷酸二氢铵缓冲液 (体积比为 2∶ 1 ,p H4) ,流速为 1 m L/min,紫外检测波长为 2 90 nm,以 SFZ-47去甲基衍生物[3 H-1 ,2 -二氢 -2 -(苯胺基 )甲基 -1 -吡咯里嗪酮 ]作内标物 .本法的选择性强 ,精密度、准确度符合生物样品分析的有关要求 ,可用于 SFZ-47在犬体内的药物动力学研究 .A HPLC method was developed for the determination of SFZ 47 [3 H 1,2 dihydro 2 (4 methylphenylamino)methyl 1 pyrrolizinone], a novel anti inflammatory and analgesic agent, in dog plasma. A Kromasil C 18 column was used and the mobile phase contained methanol and ammonium dihydrogen phosphate buffer (2∶1, V/V , pH 4), with demethyl derivative of SFZ 47 as the internal standard. The analyte was determined by UV at 290 nm. The calibration curves are linear over the range of 10~1 000 ng/mL. The limit of quantitation is 10 ng/mL for SFZ 47 in the plasma. The method has proved to be simple and sensitive with a good specificity, a precision and an accuracy. It is suitable for studying pharmacokinetics in a dog body.
