摘 要:目的 观察洛沙坦对大鼠肾性高血压的治疗效果及其对大鼠心肌血管紧张素Ⅱ 1型受体亚型AT1a mRNA、AT1b mRNA表达的调控。方法 改进传统的两肾一夹肾性高血压大鼠实验模型制作方法 ,采用定量反转录聚合酶链式反应 (QRT PCR)方法对AT1a mRNA、AT1b mRNA进行定量。结果 ①以丝线细钢丝代替银夹可以成功构造大鼠肾血管性高血压模型。②同假手术组大鼠相比 ,模型组大鼠心脏指数明显高于前者 (P <0 0 5 ) ;而治疗组大鼠心脏指数则无明显差异 (P >0 0 5 )。③肾性高血压形成后 ,模型组大鼠心肌AT1amRNA下降 ,而洛沙坦治疗后治疗组AT1b mRNA显著上升 (P <0 0 5 )。结论 洛沙坦有降压作用 ,也能逆转肾血管性高血压引起的心肌肥厚 ,洛沙坦对AT1亚型mR NA的调控差异 ,提示洛沙坦可能对AT1a和AT1b有受体选择性。AIM To study the effects of Losartan on the rat renal hypertension and the regulation of the expression of the myocardial AT 1a mRNA,AT 1b mRNA. METHODS The traditional rat two kidney one clip (2K1C) renal hypertension model was modified and the expression of AT 1a mRNA,AT 1b mRNA was examined by quantitative reverse transcription polymerase chain reaction (QRT PCR). RESULTS ①The animal model was successfully built by using silk thread and a very fine steel wire instead of silver chips. ②Compared with the rat of the SO (Sham Operation) group, the NO (Non therapy Operation) group had a marked higher cardiac index while the LO (Losartan Operation) group had an undistinguished one ( P >0 05).③The level of the expression of AT 1a mRNA of the NO group rats was decreased significantly after hypertension of appeared and that of AT 1b mRNA of the LO group rats increased markedly after Losartan was administered. CONCLUSION Losartan has antihypertensive effects and can reverse myocardial hypertrophy induced by renovascular hypertension. Losartan regulates the mRNAs of the AT 1 receptor subtypes differently, which suggests it possibly has selectivity for AT 1a or AT 1b .
