摘  要:目的:观察心肌缺血再灌注后心肌细胞凋亡与一些细胞活素水平变化的关系,以及黄芪的作用。方法:69只大耳白家兔,随机分为心肌缺血-再灌注组、黄芪治疗组、心肌缺血坏死组、假手术对照组。采用脱氧核糖核苷酸末端转移酶介导的缺口末端标记法和心肌细胞DNA电泳检测心肌组织的细胞凋亡情况,并应用MTT法检测外周血单个核细胞分泌IL-1、IL-4、TNF、IFNγ的水平。结果:缺血再灌注后,凋亡的心肌细胞增加,同时伴有IL-1、TNF、IFNγ升高和IL-4水平降低。黄芪治疗后只有少量的细胞凋亡及一些细胞活素的轻度变化。结论:心肌缺血再灌注心肌细胞凋亡过程中,细胞活素可能有重要的作用;黄芪可抑制再灌注时的心肌细胞凋亡,此作用可能与这些细胞活素的水平变化有关。To observe the relationship between the myocardial apoptosis by ischemia/reperfusion and the changes of cytokines and effects of Huangqi on it. Methods: Sixty-nine rabbits were grouped as ischemia/reperfusion group, Huangqi group, ischemia-infarct group and normal group. The myocardial apoptosis was observed by TUNEL and DNA electroprog-noresis. The levels of IL-1, IL-4, TNF and IFNr were gauged by MTT method. Results: After ischemia/reperfusion, the myocardial apoptosis was increased, while the levels of IL-1, TNF and IFNy were elevated and the level of IL-4 was depressed. After treatment by Huangqi, there were a few of myocardial apoptosis and mild changes of cytokines. Conclusion: The myocardial apoptosis can be induced by ischemia/reperfusion. The cytokines plays an important part in myocardial apoptosis. The Huangqi may arrest the development of myocardial apoptosis. This effect of Huangqi is related to its regulating cytokines.
