摘 要:目的 应用并评价巢式聚合酶链反应技术诊断肺炎衣原体 (Chlamyoliapueumoniae ,Cp)感染。方法 采用以衣原体属及Cp种的 16SrRNA特异片段为目的基因 ,对已知鼻咽拭子Cp培养结果的 81例病例进行连续 2次扩增 ,检测Cp基因组DNA ,并对其应用予以评价。结果 以培养结果为标准 ,PCR方法的灵敏度为 5 /17(2 9.41% ) ,特异度为 5 2 /6 4(81.2 5 % ) ,阳性预计值为 5 /17(2 9.41% ) ,阴性预计值为 5 2 /6 4(81.2 5 % )。结论 这种巢式聚合酶链反应技术尚不能用于临床诊断Cp感染 。Objective Application and evaluation of nested polymerase chain reaction(PCR) for diagnosis of Chlamydia pneumoniae (Cp)infection. Method Based on detecting the Cp specific region of 16S rRNA gene, 81 cultured nasopharyngeal swabs were amplified by nested PCR and compare the results with culture results. Results The sensitivity and specificity of this nested PCR was 29.41% and 81.25% respectively. While the positive value of predictive(PVP )is 29.41% and the negative value of predictive (NVP) is 81.25%. Conclusion The result indicated that nested PCR should be further improved for clinical use as diagnostics. It is mandatory for assessment of its significance with golden standard for clinical application.
